Optimal selection of monoclonal cell lines producing high amounts of biopharmaceutical human IgGs.

Jolanda Gerritsen, Technology Expert, Cell Line Development, Genmab, The Netherlands

Genmab have optimized and integrated emerging technologies to achieve an optimal, state-of-the-art process for generation of CHO cell lines expressing therapeutic IgGs. Dr. Gerritsen explains how expression levels and quality were improved and time lines decreased by implementing a ClonePix™ system to select high expressing cell lines and enable fast clonal cell line generation. She also describes the integration of a μ-24 BioReactor as a high throughput bioreactor system to mimic the conditions of a standard bioreactor.

CellReporter webinar: Accelerated high content screening and homogeneous quantification of soluble IgG levels

Dr Alasdair Robertson, Head of Biology R&D, Genetix Limited

Dr Robertson demonstrates how utilizing the CellReporter™ system's quantitative approach to assessing IgG levels can save time and simplify workflow compared to conventional methods such as FMAT®. Data was also be presented on how the high resolution imaging and cell-by-cell analysis of CellReporter benefits a variety of cell-based assays.

Phenotypic screening using CellReporter to identify cures for the deadliest poison

Dr. Tobin Dickerson, The Scripps Institute

Dr Dickerson will discuss the development of a whole cell based assay for Botulinum neurotoxin (BoNT) inhibitors based on the new CellReporterTM multi-application platform. CellReporter enabled multiplexing of their functional assay with other drug development filters such as cytotoxicity. This new approach allows them to efficiently triage lead compounds to optimize those that enter in vivo studies of BoNT intoxication.

Efficient Single-cell Cloning for Mammalian Cell Protein Expression with ClonePix FL

Dr. Cheng Liu, President and CEO, Eureka Therapeutics, Inc.

Dr Liu discusses how ClonePixTM FL has helped Eureka Technology save months of time and labor costs compared to traditional limiting dilution method. When used for sub-cloning of existing antibody-producing cell lines, they have seen improved single-cell clonality and often better expression levels.

Please note, due to technical difficulties during the webinar the sound will not start until slide 5.

The use of ClonePix FL to increase productivity in a research-based hybridoma facility

Dr. Robin Barbour, Associate Director Elan Pharmaceuticals

“We have implemented ClonePix FL to increase our fusion productivity by approximately 50%, while decreasing our time from fusion to stable clone by 50%”

“We have been able to perform primary screening of fusions directly in the initial fusion plates, thus eliminating the need for time-consuming ELISA assays. We can also assess stability, ensure clonal status and determine highest producers in a single cloning step. We have been able to rescue unstable clones and increase productivity of certain hybridomas with minimal efforts.”
